Ok I know the first slaves that came to the US were from Africa. What language did They Speak? Could they understand their Masters?

It really depends upon from where they were captured. Typically, masters forbade their slaves from speaking in their native tongues and were forced to learn english. You may find this article interesting

They probably spoke the language of their tribe or region in Africa, but they most likely learned English very quickly out of necessity.

What ever language their masters told them to speak. They were slaves.
