It depends on what style(s) of comedy appeal to you the most....
If you're the slapstick type, then I'd recommend Spaceballs, Dumb & Dumber, or Private Eyes (with Tim Conway and Don Knotts... classic).
If you're the dark comedy type, then I'd go with Little Miss Sunshine or Harold & Maude (there are more, but this is off the top of my head).
If you like British humor, then Monty Python & the Holy Grail, Life of Brian, The Meaning of Life, A Fish Called Wanda.
For romantic comedies, Love Actually, Arthur and 50 First Dates come to mind.
Others that should be a part of any funny movie list: Meatballs, Stripes, Blues Brothers, Blazing Saddles, History of the World Part 1, Waiting for Guffman, Best in Show, A Mighty Wind.
I'd try to think of more, but the baby's crying... gotta go!
