I would have to say Steve Vai. If you get a chance watch the film Crossroads with the boy from the karate kid. Steve wrote and played all of the guitar work in that film. The duel at the end is brilliant. And before anybody corrects me I know that Ry Cooder wrote the songs, but the guitar work is all Vai.

Jimi Hendrix, He played the quitar with everypart of his body at
one time or another and like it was part of his body.
He did things with the guitar like no other before or after.
Most guitarists will list jimi hendrix as a big influence in their style of playing and jamming.

Jimi Hendrix and Eddie Van Halen (tie)
Jimi could do anything with the guitar...Eddie had some help but to have it patented (the mechanism whereby he was able to hold it like a steel guitar and do hit tricks) was pure genius.
