I can't give you a straight answer. Each of the guys I am about to name gave so much to their music, and had such different styles of playing.

Brian May - Queen guitarist. No one can replicate his sound. His skill is simply amazing.

Hendrix - Completely revolutionized guitar playing in general. The fact that he sung while playing makes him even better.

Clapton - I'm not a big Clapton fan, but I'll give him credit for being one of the best ever.

The whole Eagles guitar section (Joe Walsh, Don Felder, Glenn Frey) - they really were awesome innovators and very talented musicians.

Jimmy Page - revolutionary, very talented.

Eddie Van Halen - played the dual (or whatever you call it) guitar (two necks?) which is awesome. Some of the stuff he did is amazing.

Steve Vai - I've never heard him, but he's regarded as one of the best by a lot of people.

George Harrison - He was an incredible innovator. Played many instruments besides the guitar. Listen to "While my Guitar Gently Weeps" an you'll see why.

Angus Young - completely unmatched speed-wise. Some of his stuff is just plain crazy too.

I feel like I'm forgetting someone, but I can't remember who. Oh, the dude from Cheap Trick (don't know his name) played a massive guitar, like 6 necks or something like that.

Some guys who were good, but not the best:
Slash - of Guns N' Roses
Neil Schon - Journey
Rick Savage - Def Leppard
Joe Perry - Aerosmith
The Edge - U2 - has his own style, worth mentioning, but not amazing or anything like that.

Hope this list sheds some light on the question.

I know everyone is going to be screaming Jimi Hendrix from the top of their lungs, but I have a different answer. This might not count, because he was a bassist, but Dimebag Darrel. RIP, Brotha Dime.....
