As far as the relationship between Mary Tudor and Elizabeth goes, they were siblings, but there was rivalry for the throne. Even though Mary, as eldest, was entitled to inherit first, a lot of people didn't want her because she was a staunch Roman Catholic. They would have preferred Elizabeth, so for a while, Elizabeth's life was in danger from her sister. Eventually, Mary did take the throne. She held it for five years, and earned the name "Bloody Mary" because she burned anyone who didn't adapt to her religion. After her death, Elizabeth took the throne as Queen Elizabeth I.

Mary Tudor went on to become the Queen of England, but she always lived in fear Elizabeth (or her supporters) would take the throne by force.
When Mary's father divorced her mother, Henry had Elizabeth with Anne Boleyn and May went from being the heir to the throne to being Henry's illegitimate daughter, while Elizabeth took her place. Then, Mary was forced to wait on Elizabeth as lady-in-waiting.
Throughout Henry's many marriages, both Mary & Elizabeth's positions were constantly threatened. As sisters, they loved one another, but were constantly aware that each stood in the way of the other's hopes for the throne.
It was a very difficult relationship, to say the least!

Mary Tudor was Elizabeth I's older sister. Her mother was Catherine of Aragon and her father, King Henry VIII. Elizabeth and Mary shared the same father. Mary actually wanted Elizabeth dead since Elizabeth's mother, Anne Boleyn, had been the cause of the break-up of her parents. Mary Queen of Scots was Elizabeth I's cousin. Mary was behind a couple of plots to kill Elizabeth I. Elizabeth gave her as many chances as she could, but in the end, had to have her head chopped off. James I, who became king when Elizabeth died, was Mary Queen of Scots' son.

Mary Tudor was 'Queen Mary I of England', aka 'Bloody Mary' (1516-1568). When she died, her younger half sister, Elizabeth I (1533-1603), became queen. The were both daughters of Henry VIII.
'Mary, Queen of Scots' was Mary Stuart (1542-1587), their first cousin once-removed. She's the daughter of King James V of Scotland. Her grandmother was Margaret Tudor who was married to King James IV of Scotland. Margaret Tudor was an older sister of Henry VIII.
There is another Mary Tudor (1496-1533) who was the younger sister of Henry VIII. She was the aunt of Queen Mary I of England and Elizabeth I. She was married to King Louis XII of France.

Mary Queen of Scots was daughter of Margaret of Anjou and the king of Scotland. She married the heir to the thrown of France and was queen of France for a short while until his death. She then was Queen of Scotland. She was beheaded after many years of imprisonment.
