what do you mean by biggest? the longest? the best?
if you look at what rivalry did the most for it's sport that would have to be tennessee vs ucon in womens basketball. that rivalry has brought the womens game to where it is. if you are looking for tradition in football it would have to be ole miss and mississippi state. if you want atmosphere it would have to be alabama vs tennessee nothing like the third saturday in october. and basketball there is duke vs norh carolina. so just what do you mean by biggest?

I cannot believe nobody said this: Indiana vs. Purdue!!!! Especially in basketball. When Bob Knight was at IU it got absolutely NUTS! Nutty enough that coach Knight would throw a chair across the floor! That game was against Purdue! I have noticed that when Indiana and Purdue play each other the intensity could easily match that of a national championship game.
