If it's a system file (in use by Windows) you will have to boot from a floppy, USB drive, or CD in order to delete it.
If it is not a system file just right-click the file & choose propeties ... make sure that "read only" is unchecked.
Tell us what the full filename is (with the extension) & we may be able to give you more precise instructions.

Hm. Try Restarting your computer or:
Download ViM: http://www.vim.org/
ViM is an editing program to edit the text of a file.
Install ViM.
Right click on the file you want to delete, go down to 'Edit with ViM'.
Go to 'Edit' on the toolbar, then 'Select All'.
Press 'Delete'.
Keep doing this until it is all deleted, it may take a few Select Alls and Deletes.

if it is saying is in use check that no programmes are running that may be using it press alt ctrl and delete this will show programes running sometimes if they are set they run from startup and thats why the file may be in use if that what tha problem is,do you have more than one person using your computer that may have protected a file for some reason ,check.

Make a note of the file name and location, re-boot PC in DOS mode, walk to relevant directory and delete unwanted file. This ought to be quite simple, but it seems to bcome mre difficult as operating systems "improve"!

hit alt delete and ctrl at same time to open task manager see the file or program you are looking isn't running if it is end task and then try to delete,i'm assuming you tried Start ,Settings Control panel ,add/remove programs .if still unsucsessfull
try right clicking on START and hitting explore look for the source folder maybe in programs and if you find it try right click then delete

this depends on what the file is. alot of seemingly useless files are used by the computer in the background and are vital to its running. if you post the name of the file it will be easier to identify how to delete it.

click start,click run, write in the word regedit and then trace it on the register and completely delte it, but take care this is the computer register, and any deletions will be permanent.

Change the permissions on the file.. it will be under the "properties' for that file.. uncheck "read only" and that should work.
