Sites as big as MySpace and Facebook are not created *with* software - they *are* software. They employ teams of developers that write the MySpace software in-house. MySpace uses mostly ASP.NET and SQL 2005, running on a custom implementation of distributed database hardware and load-balancing web server farms. Facebook uses PHP for the front-end, and numerous technologies on the backend. MySQL is a popular database companion for PHP, so I suspect their database systems are primarily that.
There are some tools to create social networking sites, but they are for small-scale, generic sites - not highly specific, specialized ones. If you aren't planning on having 160 million users, any of these should work for you:

You'll need a huge server, lots of bandwidth, an SDSL Web connection, ASP, PHP, SQL, templates, too much time on your hands and more.
