The command syntax will vary a bit from router to router (you didn't say what you're using) but the classic router commands for testing connectivity are ping and traceroute.

Suppose you want to test connectivity to 10.1.2.3. On a Cisco router for example, you can just type ping 10.1.2.3 or traceroute 10.1.2.3. If you have enable/privileged access, you can use the "extended" versions of these commands just by typing ping or traceroute and then hit enter. It will then go into more of a menu driven utility with lots of options.
