I am planning to drive from Denver to chicago in a day or so without turning off engine, except for gas and food
Should I need to cool down engine after driving 5 hours or 6 hours
Bcz I am driving old 94 Accord.
How long can I drive without a stop? If I drive continously will it harm my engine

If it was boiling outside, I'd be a little concerned, but just make sure all your fluids are topped up, especially the rad and tranny fluid.
As long as the temperature gauge is working and doesn't go up past 2/3rd's, then you should be just fine.

If you drive that 1000 miles from Chicago to Denver without shutting off the engine, except for food and gas, you might actually find that your car will seem to run a little bit better.

I have driven from coast to coast in old cars with multiple drivers and never stopping for more than an hour. As far as I can tell the engine seems to run better and better the further you go. I have blown an engine in Kansas, but that was because it was so old it used 5 quarts of oil between gas stops.
