I would go to Cape Town, there's so much to see but you'd have to fly from Joburg as it would take too long to drive.
The people are more cosmopolitan, a great mixture of cultures. There are loads of places to stay - try a guest house B&B as they are usually more informal, run in a family way and so you get a bit more insight into the people. See the Rooms for Africa website link below. If you want to stay right in the city then I'd recommend the Bo-Kaap Lodge, lovely place with a fantastic view of the city and Table Mountain (link below).
If you want to spend a lot of time at the beach then go to Durban with its Indian Ocean and sharks. If you want to see some of the things that are really associated with South Africa then go to Cape Town. Table Mountain, Robben Island, Cape Point, Chapmans Peak Drive, the penguins at Boulders in Seapoint... further afield up towards the Garden Route - places like Hermanus where you can go whale watching at the right time of year. You won't have time to do some of the other bits of the Garden Route but they are so worth it.
The winelands are out Stellenbosch and the surrounding areas which is about an hour from the centre of town.
Head to Strand or Gordon's Bay for great beaches (Strand beach is the best in the Western Cape, beautiful sand and miles of it) plus the water is much warmer than on the other side of the coast as it's part of False Bay.
Cape Town was voted one of the top 5 places to visit before you die!
Wherever you go, have a great time!

I personally prefer Durban way more than Cape Town, but in general CT is the bigger attraction. They are both great places, so I'm sure that whichever you choose you will be satisfied! The vineyards as far as I know are only in the Cape, but in Durban there are the sugar cane fields. The costs of accommodation will pretty much vary in the same price ranges in both places. I hope you have a great trip! All the best:-)

Definitey definitely Cape Town. I live in Cape Town but I have lived in Durban and trust me Cape Town has WAAAY more to offer. There is STUNNING beachs, the mountain, the waterfront, wine farms (vineyards). Most of Cape Town is safe, as long as you're not stupid - like walking around alone at night. Go onto google and google images from CT and Durbs, that will help you make your decision when you see how beautiful CT is... And why are you going to JHB??? horrible!

I would recommend that you dont go to the big cities of South Africa there is alot of small towns which is much more interesting and beautiful and you will get a better taste on how south africa is.
Big cities like Durban, j'burg and cape town is just like any other in the world but the small towns is what makes South Africa different

It's cheaper in Durban + your on the warm Indian Ocean there. Cape Town is nice, but it's just a city, plus Durban is a shorter drive from Jo'Burg than Cape Town is.
Don't forget to goto Sun City while you are there, that place is really neat!

I personally like to go to Durban as it is on the Indian Ocean, so you won't freeze you're nuts off. Cape Town is a very nice city, but the vineyards is in Stellenbosch area, so it is a bit of a drive from Cape Town itself and a week is a very short time to go there. If you go to Kwazulu Natal, you should go to Margate, as it is great there. You can learn to surf whilst you there, you can go deep sea fishing. There is lots of reastaurants and nice clubs and bars, so you won't be bored for things to do.

Cape Toown. there's an outstanding holiday to Stellenbosch wine turning out to be area, with ideal ecosystem and astounding previous wineries. Whale Bay could be a competent holiday too. circulate up table Mountain, remarkable perspectives. Jo'burg is hilly and has some advantageous places around, and the zoo in Pretoria no longer a techniques away is astonishing. You get a cable motor vehicle to the precise and walk down. Cape city is the terrific nevertheless, this is have been given all you point out.
