Monty Python and the Holy Grail.

Don't know if its the best but Jackass and Jackass 2 made me laugh the most. For true films it's definitely a Mel Brooks film, but hard to tell which one.

What! Nobody mentioned Monty Python's 'Life of Brian'. Any film with lines like "He's not the Messiah, he's a very naughty boy" or "Did he say 'Blessed are the cheesmakers?' Speak up we can't hear you" has got to go at the top of my list.
