I told my assistant to get to work and she got mad. What would you do?

Depends on how you stated it and what she was doing at the time. Find out why she got mad, if its just because you said something to her if she was slacking off - ignore her and keep the idea a looking for a new assistant in the back of your mind. In the future if you see someone not doing their job, casually ask them what they are doing and then assign them some type of task.

Was she on the clock? Was she not doing her job? Maybe you could have put it more gently, but as long as you answered yes to the previous questions, you had every right to tell her to get to work.

Maybe you need to reconsider her suitability for employment with you.
