Enzyme is very sensitive to the pH of the environment. If the pH is not well controlled, then it may lose/reduce its activite. In order for enzyme to work, you must give it the right pH and temp that it "likes".

It can cause protonation or de-protonation of the side chain groups resulting in a change, or "denaturation" of the enzyme. It can also affect the ionic charge of some side chain groups.
