Ellos saben como jugar el partido. You would use saber instead of conocer because saber is knowing HOW to do something, such as playing a game, while conocer is used for knowing people, places, etc. or being familiar with them. 'como' is how and then you would keep jugar in the infinitive form because it is TO play. Then, el partido, the game. Good luck!
