Try looking at these websites. These explain procedures that can be used to extract DNA from a cell. The first web site should give you a basic understanding, and the second is a pretty simple procedure that can be used in sterile lab. The other two may help you to better understand(the last is a PDF, so it may take a minute to load). These sites are mostly from when I had to design a lab to extract DNA from E. coli cells.
