I presume you mean a metal, weapon intended for stabbing and slashing, not some phallic symbol for something else.
The largest sword in comics was the one being forged by Surtur when Walt Simonson took over Thor; it was enormous, much larger than any human could have wielded. Surtur was a fire giant/ demon, so he had the size to grip it and the muscles to swing it around. That was right around the time that Beta Ray Bill was introduced.

Not a video game, but in Michael Moorcock's "Elric" stories, Stormbringer is supposed be five feet long. That's pretty long for an iron broadsword. Definitely a two-handed weapon!
