New York and Las Vegas would definitely be on my list. Both are unique and fun. Orlando is a bit more dubious, but I suppose it's alright, if you love Disney and can't get your fix elsewhere.
Just south of whitebread Orlando is the awesome city of Miami. It's like being in Cuba without the dictatorship.
The "big three" cities in California (the nation's most populated state) are all worth a visit: Los Angeles, San Diego, and San Francisco. They're wonderfully diverse, and each is beautiful in its own way.
Honolulu in Hawaii is an interesting mix of Hawaiian, Japanese, and Filipino cultures.
Chicago is a surprisingly friendly metropolis. You should visit if you happen to be in the Midwest.
Those are the "must-sees" on my list, although there are any number of other cities in America that are interesting (Austin, New Orleans, Minneapolis, Seattle, Portland, Boston, Philly, DC).

I highly recomment Santa Fe, New Mexico. Very safe. Good if you like art and museums and history and beauty.
Estes Park, Colorado is beautiful in the summer. (beautiful in the winter too, but very cold) There are multiple mountains over 14,000 ft. Hiking, fishing, horseback riding, shopping, camping, beautiful scenic drives, cool summer weather, fun for families.
Seaside, Florida is a great place to go in the summer. It is a small beachside community that is the safest I have ever felt. Shopping, dining, wine tasting, sugar-white beaches, beautiful blue-green water, outdoor movies for the kids in season. Very clean and classy place.
This is a very small town, but I always have a great time. Especially in the spring and fall when the crossdressers hold a convention. It is Eureka Springs, Arkansas in northwest Arkansas. It is a small victorian town with plenty of shopping, good food, simple entertainment, always some sort of parade on the week-ends. B&B's are everywhere as well as spa's, ghost tours, horseback riding, fishing, canoeing. Everything is uphill or downhill. Very pretty area.

Cincinnati, Ohio. It is just amazing with all of the culture and entertainment. Any California city as well, including Los Angeles and Hollywood. Chicago, Boston, and Atlanta are some famous ones as well.
