It depends. Many of the major high street Post Offices have a Bureau de Change buying and selling foreign currencies including the dollar. However, your local sub-post office will not offer this service.

Do you mean you want a 'hundred dollar bill' in UK funds, or in US funds? If you mean in UK funds, then that depends on what you want it for ... and I'd say that going to a bank is a better idea. If you want it in US funds, then you need to go to an 'exchange bank' (one that 'changes currency' for people along with other 'banking') to get that one hundred dollar bill.

Only larger PO have exchange facilities. The large main Post Office in your nearest town should have one.

$100 are the one type of note most often counterfeited, so probably not, assuming your PO deals in Fx. Lots of places won't deal with $100 bills, much to the dismay of American tourists who assume they can change them at banks on arrival.
I live in Ireland - I'm guessing it's the same in the UK.

The post office? Is that where your bank account is?
Well, if such bills are being printed (if not, they probably will in 10 years' time), then you can get them at your bank.
