How would I set up and carry out an experiment to extract DNA from a kiwi fruit? Thank you to anyone who can help!

Assuming you're working from home:

1. Disrupt the tissue (mix up a saltwater solution, with about 3 tbsp of salt in a pint of water, put equal parts saltwater and kiwi in a blender and blend. Add enough saltwater so the mixture can be poured into a cup.)

2. Break up the cells (add a shot of clear dishsoap to the kiwi/saltwater, mix well, this disrupts membranes)

3. Get rid of some proteins (add a dash of meat tenderizer, again mix well)

4. Precipitate the DNA (Pour a little of the soap/salt/kiwi mix into a clear tube. Add cold alcohol (rubbing alcohol is fine) to the top of the tube slowly. The DNA will begin to "bubble out" of the bottom layer as a white cloud.)

It is necessary to cut and blend the kiwi to segregate the kiwi cells and break/lyse the cell. This is to allow DNA to be outside of the cell. DNA are polynucleotides while histones are polypeptides. When histones are removed, it will increase the purity. In other words, we are separating polynucleotides and polypeptides. DNA are coiled around histones. When you separate histones from DNA, it will increase the purity because the DNA will be free from proteins such as histones. A nucleus only means that DNA is surrounded by a membrane (nuclear membrane). A bacteria do not have nucleus, it only means that it do not have a membrane which surrounds the DNA. A bacterial DNA is circular and localized in the bacterial matrix. Its known as nucleoid. Bacterias even have other DNAs known as the plasmids.

something to do with mashing the pulp up to extract the juice ~~ adding ice cold ethanol to it ~ wait for some kind of precipitation ~ find thin spindly DNA ~ extract with tweezers *this part is hard* ~ have fun!
