In the original cartoon - by a chemical spill - I think with a lightning strike at the same time.
Barry allen, the scientist, got covered in the chemicals and 'zapped' by the lightning. After which - he was "Flash".

As noted in a previous answer, there have been several Flashes...
Flash I (Jay Garrick) - gained his powers through an accident while he was experimenting with "heavy water"
Flash II (Barry Allen) - gained his powers through lightning striking a rack of chemicals in his lab.
Flash III (Wally West) - gained his powers the same way...lightning striking a rack of chemicals
Flash IV (Bart Allen...aka Impulse) - descendant of Barry Allen from the future...his powers are genetic if I'm not mistaken.

The first few answerers basically had it right. Jay Garrick got his speed through inhaling heavy water fumes, Barry Allen and Wally West through being struck by lightning while doused with chemicals. Apparently, this activated latent metagenes in their cells so that their descendants (like Bart [Impulse] Allen) also had super speed.

Because he once appeared in disguise at a gathering of his Rogues Gallery, in order to recover some of their hidden stolen loot, and he called himself—The Dude! it depends on which flash your talking about becauser there are many flashes and they were all created differently.....one was struck by lightning another durin an experiment...etc...so be more specific
