You are given a broth culture that is growing an unknown organism. What is the first test you do and why?

gram stain the broth, and see if it's gram neg or pos, bacilli or cocci, or a mixture of organisms. then plate the broth to blood agar and chocolate agar, and macconkey's if you have a gram neg rod. if the specimen is from a wound or a sterile body fluid, be sure to incubate a blood agar plate anaerobically to rule out anaerobes. after that, you'll be able to narrow it down and test for the specific organisms that have the characteristics you find.

Attempt to identify the organism. Look for important classes of characteristics such as:
size
type of margin
colony elevation
colony texture
light transmission
colony pigmentation

These characteristics will help you identify the type of organism you are dealing with.

Make a slide and stain it and view it. Is it gram neg or pos. What is the shape of the organism?
