I recently filed a business name, i'm using it online as a domain as well. I discovered that a well established co. uses the same name; it only differs by 1 letter. We do not sell the same products but are in the same industry. Can they go after me?

Are you deliberately misleading consumers? If not, I don't think the other company can do anything about you have a similar name. In the physical realm, there are so many companies with similar or imitation names it's not funny - Dairy Prince, Dairy King, Dairy Duke, Dairy Jr - just to list a few knock offs of Dairy Queen

I think you can use it, obviously- you registered the domain name and the fictitious name with the State. If the other Company has a problem with you using "their" name- they could file a lawsuit against you for some sort of identity infringement. If your intent was to make profit "on their" name and they can prove it, they may prevail via lawsuit against you for damages or injunctive relief, etc. Best to check your state laws on name usages-

Technically, if your name is different even by one letter or number, you have a legal right to use it. They may come after you because they don't want the confusion of having another company that is so similar.

Yes they can and more than likely will. There was a shop called Blinds R us. They got sued by Toys R Us and lost. You need millions of dollars to figtht the BIG guys.
