The answer may be a little more complex, depending on what your specific circumstances are.
Basically, yes you can use more than one iPod with iTunes.
If you use a Mac, you can connect more than one iPod simultaneously.
However, if you are using iTunes for Windows this is not supported (it may work, but you may experience 'unexpected behaviour').
If you are using Windows you can still use more than one iPod with iTunes: you just need to only have one plugged in at a time.
When you plug an iPod into your Mac or PC, iTunes can be linked to your music library. iPod is synchronised to the contents of that library of music, so if you delete music that you don't want on the first iPod, you won't be able to sync it with the second iPod.
The best way to have different music on different iPods is to either use different user accounts on your computer (therefore different iTunes libraries) or to create specific playlists which you tell each iPod to update from when using one account only.
As always, one of the best places to look first for iPod info is Apple's Support site at www.apple.com/support (click through to iPod + iTunes). Also try the discussion forums there, as well as places like iLounge (www.ilounge.com) and MacRumors forums(http://forums.macrumors.com/forumdisplay.php?f=75).
I hope this is helpful : )

Yes.
You can sync them both to iTunes.
If you want different songs on them that is different.
I don't know what to do if you have a Windows computer but if you have a mac you can download multitunes of of the apple website.
Sorry 'bout that if you have a computer running on windows.
