Well there aren't very many athiests nowdays anyway. Most people who considder themselves to be atheist don't really understand what athism is and they are actually agnostic. The high number of agnostic people shows that many people don't even know what their true calling is, but they believe there is a true calling.
To answer your question: If, as atheists believe, that we're all just here by chance, then one knows they've found their true calling once they have discovered what their true calling is.
People use 100% of their brains.

Maybe you are more confused than the atheists for they know what they're talking about but you on the contrary leave your question unfinished as if by chance the reader will be able to read your mind.
OK. We are not here by chance but by just a coincident. What I mean by that is that when the right circumstances are created things happen and will continue that way indefinitely. If there was a god then there must be someone to create him. Isn't it?
So are you trying to tell us that everyone here knows what their true calling is? I don't know my "true calling" Do you know what your true calling is other than asking a dumb question like this?

No one can 'know' their true calling - if they believe in that idea. People can only 'think' they know.
An atheist can still believe in a true calling because they may feel their talents and desires are best suited to certain activities.

How do non-atheists know?
Not like God(tm) calls you up and says " Hey, dude, being a plumber is your calling"
You know when you feel right about what you do, when you like getting up to go to work, when you find ways to learn more about your career than simply what's needed to do the basics of that job.
Also, when you're good at your job...

first of all...not all "atheists" believe what you mention.
What do you mean by "true calling". My profession? My destiny? What color shirt I will wear a year from now on this date?
If you worry yourself with your question, you will find that life just passed you by.

I like spinoff and Richard A. too. Good answers.
We are here by Divine chance, not by accident; and, if we can discover any one of our top five callings we are very lucky!
With love,

Life has meaning because we give it meaning. I decide, as best I can what my calling is.

I asked God, and he said my "true calling" was to tell you to "shut the hell up."
