It depends if you've been there or not, what sort of things you like, and how long you'll be there, but here's my abbreviated list of places that I like to show my friends:
Chinatown (make sure you hit the shops, the garden, and have lunch)
Gastown
Granville Island (buy some fudge and/or smoked salmon)
Museum of Anthropology (at UBC, then go for a walk down the cliff to Wreck Beach)
Robson Street (hang out, see the famous Starbucks corner, shop, have Japanese food)
Granville Street (avoid the porn shops, just go for a walk)
Canada Place (great view of the park and inlet)
YOU MUST NEVER GO TO VANCOUVER WITHOUT SEEING STANLEY PARK!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!
Stanley Park! You can easily spend a day just walking around. Bring a camera.
Victoria for a day-trip. If you're spending a week plus, you can't miss this either.
I could go on and on. That should be enough to keep you busy.
Make sure you eat some B.C. rolls (type of sushi), smoked salmon, and have a good coffee somewhere like on Commercial Drive. Starbucks is fun to look at, but Vancouver is a great city for coffee and a few non-chain stores still exist and have character.
