Guitarists: what is the difference between tuning and intonating?

Tuning is the guitar in tune with it self with the strings. Intonation is the single string by itself in tune with itself. Like it should be able to have the same tone on the 12th fret as it does with its open harmonic at the 12th fret. That is the way I read it..

tuning gets you to be able to play in a better tone and intonating doesn't matter that much
