This is all about personal opinion, isn't it? The answers so far seem a bit tilted toward the rock world, but there are plenty of outstanding guitarists in other areas of music.. Chet Atkins, Les Paul, etc. My own opinion is that Eric Clapton deserves to be on all the lists of best guitarists... as does Leo Kottke. My favorite to listen to at the moment is probably Mark Knopfler.

Holy cow, you guys never get that there's no "best" because they're too different to compare. So obvious...
Django Reinhardt (ease of play, melody)
Robert Fripp (technique, innovation)
Hendrix (raw energy)
Vai (technique, speed)
Satriani (technique, speed)
Pat Metheny (everything - the most complete and amazing guitarist I can think of)
Al DiMeola (precision, speed) - kinda like John McLaughlin but with no pretense and arrogance.
Clapton, B.B. King (emotion, subtlety)
SRV would be among Robert Crays and other C-leaguers... nothing original about him.

WOW tufff question. This is hard to answer because of the different styles of playing but I would have to say David Gilmour of Pink Floyd, he is amazing.
