Because mice do not live as long as humans, they scientifically increased the level of carcinogens in the air to match a person exposed to second-hand smoke.

They autopsied the mice and looked at their lungs and blood work compared to the controlled group of mice.

Probably did that with rats. But most data was derived from getting info from people with typically smoke related illnesses, who were not smokers themselves, but lived with or worked around other smokers. After controlling for other factors, there was a clear association between their proximity to cigarette smoke and their illnesses.
