if this is a new bike, if ur dealing with harley, u most likely wont be able to go below msrp. if used, i'd have a bike mech go over the scoot from top to bottom, and see if he has any verifiable service records with it. if the ad says ''10,000 firm'', then there is no negotiation.
those who own harley's will most generally take very good care of their scoot.

First of all, a used bike should be inspected by a dealer before you buy. Do a search and find out the price for the model and year of bike that you want.
As for negotiations, go around the bike and find everything wrong you can about it, blue smoke, scratches, etc. Start low and keep your maximum in your head. Do NOT go over your maximum. Ck the tires, brakes, etc...

If you don't really know what you are doing, only buy a new bike from a dealer. You have some protection there.
Only buy a used bike from someone you know or trust, or have someone who knows motorcycles well, help you inspect the machine in question.

Get a motorcycle endorsement first. Take the course and make sure you know how to drive a motorcycle.
