Railroads gave rise to big business in two ways. First, by speeding up the movement of goods and lowering transportation costs they created larger markets. This in turn allowed for large business involved in the mass distribution and mass production of goods. Second, the railroads were themselves the first modern corporations. Their model of corporate organization was imitated by businesses (and non-business entities) throughout the economy.
Railways were a force in the closing of the frontier by promoting settlement,making farming economical on the prairies and by their role in the slaughter of the buffalo herds & otherwise facilitating the removal of the Indians as an obstacle to settlement.

Yeah its me again.
I don't know that Rail roads did give way to big business. They may have helped bring the two coasts together (close the frontier) and so enabled coast to coast business start and develop.
With the development of the railroad you could then travel from east to west in days rather than months. This is good for business - communication and moving people around.
If you like something you take it with you. So there is a service/product you like as people moved inland they took these with them and perhaps enabled bigger business to develop.

Another factor involved with big business is finance.
How did they get the money to start out?
That would be something to look into..... Follow the money trail.
