Wow ! What a great question. It requires some real thought.
Certainly, for just her popularity over half a century, you have to put Lucille Ball near the top if not actually number one. She was number one for so long that we tend to forget that longevity. But she was not particularly successful in her films, and her nomination for an Oscar for "Mame" was a courtesy only.
All the other film actresses I can name were successful in both comedy and serious dramatic roles, but would not be categorized as "comediennes". Doris Day was wonderful in a comedy, but she did some great dramatic roles as well. She was one of the biggest box office stars for more than 10 years.
Jean Arthur, Shirley McLaine, Katherine Hepburn, all did great comedy roles, but were better known as dramatic actresses.
Nope. I can't come up with a better name than Lucille Ball right now. On television she was unique. I suppose for her longevity over many different series, currently Mary Tyler Moore was very good. Going back to radio, Gracie Allen was the most popular comedienne of her era of the 40's and early 50's , but she retired because of ill health, and is not remembered by another generation today.

Alive: Wanda Sykes is like a female Chris Rock and the funniest woman i've ever seen.
Passed: Lucille Ball was a brilliant comedien and her old show is still as funny today as it was back in the 60's.

Phyllis Diller ! She recently did a Stand-up Comedy Act on the Tonight Show, and She was downright funny ! So, why is this significant ? She is about to turn 90, and has Promised to make another Appearance on the Tonight Show on Her Birthday!. Now, the Question was "In History". So, I ask You, who has more of a Claim to Fame that spans nearly a Century ? Wow ! I thought She was Dead ! Didn't You ? And She still looks and sounds about the same !

I Love Lucy - she was great and paved the way for women to be allowed to be funny. Arguably, Carol Burnett is the best with her variety show. She could do it all.
