It is probably some sort of moss or, more likely, lichen, that has started growing. These sorts of plants grow naturally in very harsh environments (on barren rock faces), so roof tiles are a natural place for them. You could try treating the surface of the roof with some sort of herbacide or other chemical to kill it off, just be careful that it doesn´t get washed down the gutter and into your lawn next time it rains.

If it were me, though, I wouldn´t bother. Moss and lichen are harmless, and it´s not as though house guests are going to be looking at your roof.

I have been told it is a type of mold and can be killed with bleach, but bleach can weaken your shingles. I don't like the way it looks but haven't been able to wash it off.

these blotches occur in many parts of the US.......here in the south it is on many houses..the main cause that we have found is major temp changes in the attic...check to see if u have good attic ventilation and if ur heating system is in the attic that it is not leaking in any areas.....u can power wash these areas and get most of the stain off....u need a warm water washer , u would do better to hire a steam/power washer company to do this.....
