Depends for how long you plan not to use it.
Frequent On / Off or better put: frequent Hot / Cold does more damage to a PCs hardware than leaving it on (except for the monitor which should be off or on standby if not in use).
I've had my PC on for over 6 months now ... never off.
In a healthy, well ventilated, non overclocked PC leaving it on is not a problem.
Make sure you have a reliable surge suppressor.
An occasional reboot (for software reasons) is recommended.

It depends how long it will be until you use it again, and how old it is and how well ventalated it is.
If you will be using it again in a few hours, you can just leave it on. There is no point is shutting it down. Just make sure you either have a screen saver or you turn your monitor off.
If it is older, you may want to shut it down if it doesn't have good airflow to it. Computers run very hot and can overheat and things can get damaged.
If you are going to be gone for a work day, or overnight, you can use standby mode. On Windows XP, you go to start >shutdown, there you can choose standby. This will allow the computer to almost power down, and appear to be off, until you move the mouse (desktop) or hit the power button (laptop). The computer will then resume where you were when you put it on standby. Think of it as kinda a "stop" like on your DVD player.
If it going to be more than a day before you use the computer again, I would shut it down.
Make sure if you are leaving it on standby extended periods of time, that every couple days you actually shut the computer down and reboot it so any updates that run on startup can run, and that you clear the memory so it doesn't slow down a lot.

30 ~ 60 mins leave it, 1 ~ 2 hrs leave it on standby mode, 2 & above turn it off. This times are good from 28 ~ 35 Celsius

It is easier on the unit to let it go into sleep mode and just leave it on instead of starting up and shutting down all the time. However, I've noticed that my electric bill is definitely less when I shut the PC down when not in use. Even in sleep mode, the fan is running and sucking up juice.

Use your common sense... If you're leaving for 10 minutes, leave it on - if you're leaving for 10 hours, turn it off....

i am very picky about my electricity and i always turn it off whne I don't use it and unplug it...b/c even though its on standby it's still using electricity...i use it then turn it off and then whoever wants to use it again can turn it on again...
