From Mangalore come to either Tiruchirappallie or Nagercoil and change over to Tuticorin.

There's no direct train from Mangalore to Tuticorin. However, you can make two possible connections.

The first option is to take the Mangalore Tiruchchirapalli Express train (number 6868) from Mangalore (Departure 6:40am) to Tiruchchirapalli (Arrival 9:30pm). Then you take the Chennai Egmore Kanniyakumari Express train (number 2693) from Tiruchchirapalli (Dep 11:50pm) to Tuticorin (Arr 6:15am next day). Total trip is 23 hours and 35 minutes including two hours and 20 minutes connection.

The second option is to take the Mangalore Yesvatpur Express train (number 6518) from Mangalore (Dep 7:30pm) to either Mysore (Arr 4:30am next day) or Bangalore (Arr 8:20am). Then you take the Mysore Tuticorin Express train (number 6732) either from Mysore (Dep 6pm) or Bangalore (Dep 9:20pm) to Tuticorin (Arr 10:30am second day). Total trip is 39 hours including connection in Mysore for 13 hours and 30 minutes or in Bangalore for 13 hours.
