LOL, just kidding...
Actually, your best start is not to start out as a mystery at all. Just tell a story and let the mystery develop... It can be murder, a missing person, etc... You could start it with the missing person waking up, trying to remember how he/she got where he/she is, and then trying to figure out the who/what/where....
Could be a case of a stolen dog bone... Make it a comedy-mystery... Whatever pops in your head...

I always liked to walk in the old cemetery. It was interesting to see the names that had been in town for generations. I remembered coming here as a child with my mother and grandmother to tend to the family's graves. I would fetch water for the plants they brought to lovingly plant around the family headstones in the spring. Later we would stop for lunch at the town's only diner where I would get a sundae for being such a "good helper" at the cemetery.
Grandmother and mother were gone now. I had promised to look after the family plot as they had in their time and I had just finished weeding the last stone. As I stood and stretched, a peacefulness filled my being. So many stones around me were so old and many of them unreadable. As I scanned the stately rows of stones I noticed the old Derrums' family stone looked different. What was that line of whiteness on top? It didn't look like flowers. Maybe it was garbage left by kids or even a dead seagull?
Thinking to right whatever was wrong I walked casually over to the large black granite stone and gasped in shock as my eyes focused on the top of the stone.
It was a human arm severed neatly at the shoulder! No blood decorated the end and there was a gold wedding band on the ring finger!
Backing up a few steps I turned and fled to my car. The sheriff would want to know about this immediately!

and intresting way to start a mystery story is to start with part of the climax of the story. then you go back in time and end with the climax again. it's called running your story full circle.
