Singapore Discovery Centre is the first stop for a great visit... The children will learn quite a lot of interesting facts there, also Singapore Jurong Bird Park, Zoological Garden are also great. At night maybe they would like to try some night safari. Sentosa Island also has the Underwater World..
For multicultural experience, go to Chinatown, Geylang Street,Little India, Bugis Junction..there they will learn different cultures in Singapore live in harmony...
I dont recommend museums though, it might be too boring for them and difficult to understand. Bring them to an outdoor spot instead where it offers both recreation and knowledge.

Well since you are looking for a cultural theme, you can go to Kampong Glam, Little India, Geyland Serai, Chinatown and dun forget the Singapore Muesum and the Asian Civilisation museum tose tie up nicely to your theme. And if you want the children to have a bit more fun and something more singapore and a bit less cultural you should go to the Singapore Discovery Centre they have the hstory of singapore with a bit of singapore culture thrown inside.
