Hardness is the easiest, but a little training and you can tell by looking closely. Diamond has a high dispersion index.

To check for yourself, use hardness. Diamond is one of the hardest substances there is, so it will scratch almost anything. A glass plate or mirror is commonly used since glass will not scratch glass. But it will likely take an expert to tell the difference between diamond and silicone carbide, which is sometimes used as a diamond substitute and has characteristics much, much closer to the real thing than cut glass does.

Diamonds can cut class. Glass can't. Besides, jewelers can just tell by looking usually - if you look at it enough, glass doesn't really look like a diamond.
