It would be most advisable that the first time that word appears on your resume, it is spelled out in full. You can do something like this:
... Assistant (Asst.) ...
and the next time, you'll just have
...Asst. ....
From personal experience with reading resumes/hiring people, I've found it really doesn't look great if you are reading a resume and the person hasn't bothered to put the word out in full. It'd be better to alter the font, as the answer above says, or possibly to cut out some other words from that line, rather than use an abbreviation without first writing it in full.
If there is ABSOLUTELY no way to do that, then using the abbreviation isn't the end of the world. It doesn't mean you'll be rejected outright or anything, just that the resume may not have the icing on the cake.

NO, you should always spell the word Assistant' out, as you should spell EVERY WORD in your resume out. If you 'don't have room' then you need to go over your resume, word by word, and take out the ones that really don't 'matter' ... think of the old ''Dragnet" show and put in 'Just the facts' and you'll have a much better resume. I know that it's not 'fair' that good resumes get the better jobs, but if you can't 'do it yourself' then the money spent to have a PROFESSIONAL resume writer do it for you is well spent, because you'll get a better (and probably higher paying) job from it.

You could, though try to fit the whole word on there if possible. Try changing the font size to 1 smaller, sometimes that helps, like if you are typing in 12pt. change it to 11 or 10pt. I believe if you had the choice to make your resume neat or to write out the full word, I would make it neat. They will see how it looks before they see what exactly it says.

Yes, I've seen that done before. Especially when it comes to resumes, if something like that won't fit, I think there shouldn't be any problem with that. You may, however, also want to put the period after it to make sure it looks more grammatically correct.
