I know opaque means can't see through, but what does it mean when someone within the arts says, make sure you use opaque colors?

That the medium is not transparent - so the other colors don't pollute your top color by showing through.

are you talking about screenprinting??????? you need to use opaque colors in screenprinting when you create your transparencies. opaque color is one that can't let the light through. because in screenprinting, you need to expose your screen to light, anything with opaque color will be your image, it won't let the light through, and so it will let your paint through. I don't know if that's what you're looking for :) good luck
