God, what a question, Greece is crowded with things to see and do!
- Athens, naturally, a big metropolis with a nightlife, several museums, the Acropolis and the Plaka neighbourhood below, etc.
- Thessaloniki, the second biggest city upnorth, roman vestiges and a nice place too.
- Delphi, well, where the oracle was.
- Olympia, where the Olympic games took place, so beautiful in spring!
- Corynth, roman plus venetian ruins, very interesting.
- Nafplio, lovely town with a venetian fortress.
- Tyrinth, more ruins, said to have been built by giants.
- The Meteora, churches on top of rocky formations, I'm sure you must have seen them in a James Bond movie.
- Crete, the biggest island in the Mediterranean, amazing shoreline, plenty to see and do and don't forget to do the Samarian Gorges, a 16 km long canyon with a Cretan resistance (to Turks and Germans) village in the middle.
- Rhodes, a beautiful island, big as well, with lots of history.
- Santorini, the most beautiful volcanic island and it entitles you to see some Minoan ruins as well.
- Samos, where Pithagoras was born and you can go from there to Turkey to visit the ruins of the amazing ancient city of Ephesus.
- Corfu, on the Italian side, most beautiful island.
And so much more, so much more, so much more... Hope you have a pleasant stay even though you won't be able to do all that in just a week...

Greece is the "Largest small country in the world". Henry Miller said that, not I and Henry made a very accurate observation.
Greece has gorgeous Islands and they are different kinds of islands as well. Over 3000 of them. The mainland has some truly indescribable beauties, too many to mention. What you have here as comments are everyone's personal favorites and what you need to do is to get a picture book from your library, leaf through it with a map of Greece next to you and see for yourself what places you want to visit.
Friends of mine go to Greece once every 3-4 years and create a different itinerary each time. Meteora, Santorini, Meganisi, Corfu, Rhodes, Halkidiki, Crete, Kos, Delphi, Nafplio, Monemvasia, Ksanthi, the Evros Delta, Gianena, Pilio, Lake Plastira, Galaxidi, Hydra, Zagorohoria, Thasos....
How is this for a beginning?

If you're looking for an island (which I would suggest), I recommend the island of Andros. It has been a couple years since I've been there, but if I remember correctly it is a two hour ferry ride from Rafina. I would recommend staying in the town of Batsi - it has a very nice beach, couple of restaurants, and I think only one or two clubs. The people are extremely nice and the landscape is beautiful. If you're able to travel around the island (there are places to rent scooters), there are many beaches, archaeological sites, waterfalls, etc. to be seen. Plus, you can island hop via ferry from Gavrio, which is the main port on the island.
Below are a couple of links that should help you out:

http://www.andros4u.com - this is the place I stayed while visiting the island. They were absolutely wonderful and very hospitable.

I did a Contiki tour of the Greek Islands but I am positive that other tour companies also organise this. It was the most amazing cruise - Mykonos, Kusadasi (do not miss a trip to Ephesus - I know that this isn't Greece but still totally amazing), Patmos, Rhdoes, Crete and Santorini. You can't go to Greece without going to Santorini and Mykonos - two of the most amazing places in the world - absolutely gorgeous - and don't miss the sunset in Santorini - I had tears in my eyes. I know that most people don't like Athens because it is dirty, but I found it amazing. So don't miss it!

Santorini. It has amazing sunsets, different colored beaches, archeological ruins, and a cool volcano to explore. If you take the ferry from Athens there are hotel owners at the docks that have pictures of their hotels. We stayed in a nice hotel with a pool and a view for about $12 per person per night if you can believe it. And it wasn't trashy like you might think.
Athens is worth seeing for it's ruins and museums but it's trashy and polluted.

I would recommend all the islands of course! But try to visit Mani, in Southern Peloponnese! It has great places, beautiful&clean beaches! Breath-taking scenery! Especially Karhamili! It's a beautiful summer resort! It's a small village next to the sea, in the foot of Taigetos (the bigest mountain in Peloponnese)! It's very popular around Europe!

Go to Santorini. It's one of the most popular destinations in Greece. It has a breathtaking view of the blue Aegean Sea and the volcano.
As for accommodations, you can check out Panorama Studios, which is located at Fira (the city capital of Santorini).

a very good idea is to go to the Ionian islands which have some of the worlds best beaches. For example Porto katsiki and egremna beaches in lefkada or lefkas island.
And the "shipwreck" beach in Zakynthos island
