is there any difference if the saddles are not adjusted to get the right intonation?i just learn how to adjust the intonation of my guitar and i already did but i want to know,is there any difference if we don't adjust it?can someone tell me?

If intonation is not adjusted correctly, your guitar may be in tune on the open strings, but when playing notes further up the neck, they may be slightly out of tune. Sometimes it can be off very slightly, but other times it can be noticeably out of intonation.
Since you know how to adjust it, it's a good idea to check the intonation periodically. Also, old, worn out strings can cause a guitar to be out of intonation.....it's best to re-check after a string change.

The saddle is more of an adjustment or something to tweak to get fret buzz out of your sound. The amount of adjustment in the saddles has limited effect on intonation. You have as much control by bending the strings. The saddles will affect the action, the height of the strings off of the fret board. Great saddles will also allow you to adjust string spacing(distance between the strings). By moving the saddles forward or backwards, you can adjust the intonation but you really aren't typically given that much adjustment space to alter intonation much. Tension is another aspect of your stings that can be adjusted by the saddles.
