speak English , if used like this : Speak English ? It means can you or do you speak English language.
Second one means:
it is a strict way or a compulsion put on someone to converse in English assuming before hand that the person asked to do so already knows the language.

That would depend on the content of the sentence and conversation.
The correct way is: "Speak in English" if one were speaking another language at the time.
If however you're taking the English language and tearing it up with slang, than it would be a friendly non-conformative way to say Speak English (will ya)

Actually both are correct and are used in completely different situations.
'Speak in english' is used when you are askig some one to act in the desired way.That is when you want some one to use english language for speaking.
'Speak english' is used when you want to describe the way of speaking.That is if you were to express that you used english for speaking or know spoken english then you use 'speak english'.
