No- unless it's a very short one.
An average DVD holds about 8 gigs or 8,000 megs of data. A 256 meg memory card can hold about 7 minutes of high quality video or about 15 minutes of low quality video. When memory cards go above 8 gigs of memory, then a standard feature length DVD would fit on them.
By the time that happens, though, the PSP will be obsolete and maybe the PSP2 will be out and Hi Def will be the standard, for which you would need an 80 gig card for a feature length Hi Definition DVD.
At this rate, the memory cards will never be able to catch up to the amount of memory required for a full length movie.
The discs the PSP uses is a high compression, duoble layered DVD format unique to the PSP.
However, if you are referring to the TV commercial where the guy is following clues on a PSP memory card to where his girlfriend is, those are short (less than a minute) clips stored on the card.

Well first of all, it depends on the size of your video file, If your using a PC find the file on your hard drive, right click and select properties, it should tell you how big the file is. If your thinking of putting a mem stick in your memory card reader, and then putting it in you PSP then STOP NOW you could ruin your PSP- you need the PSP USB cable. An easy way to streamline the video transfer process is to go out and buy the sony media mamger software. it's only like 15 dollars and it is very useful for transfering various media.- You can also just call sony and they'll walk you through the process- they have a GREAT CS line. Yes! Go to Google and find DVD Shrink 3.2 (its free) Shrink the movie down and you should not have a problem. Even better is a program called PSP Movie creater. (not free) Bet it works very well and is worth the money. You can also use x-oom movies on PSP. (not free)
