Cool question!
For most people, the answer to that will change many times over the course of their lives. At 5, your birthday and Christmas may do the trick, and at 16, the days may be when you get your driver's licence, and your first real girl(or boy), says they love you.
At 25, it 's liable to be the day she(or he) said yes, they would marry you, and the day your first child was layed in your arms.
One thing that tends to be constant for most people, is that whatever defines those two days, the older you get, the more you celebrate FOR the ones you love, and less about what it is on a selfish level, just about yourself.

The day my daughter was born was the greatest day of my life. The day my grand-daughter was born, 27 yrs. later, was the 2nd greatest moment of my life.
No other experience has ever matched the excitement and thrill of those moments. But I have had a zillion great moments too...

The 2 greatest days of my life are the stolen works of my hardship creation which benefits estimated 30 millions people around the globe. Somewhere out there, those corrupted and dirty rich, the famous and the powerful one knows about it...

The greatest of my life is when my darling starts to feed and support me as Gigolo who told my governments that he is proudly feed and support by my gf! hah...Why? First, let me tell you the joke I heard about this. The best 2 days in a man's life are the day he buys a boat & the day he sells it. But seriously, When my daughter was born & she was healthy & my second divorce. (He was abusive).
