FIRST - it WILL NOT clear the hard drive.
To convert FAT16 and FAT32 volumes to NTFS, use Convert.exe from the command line. The syntax of Convert follows:
convert volume: /fs:ntfs [/v] [/x] [/cvtarea:filename] [/nosecurity] [/?]
To understand it all, do HELP on the convert command.

Agree with Singbloger,
It will not format the drive. However, I believe you can do this without so much difficulty in the Disk Management UI.
Control Panel -> Administrative Tools -> Computer Management -> Storage -> Disk Management.
My drives are all NTFS so not sure, but I"m fairly certain I've done it through there before. You can right click on the drive or on the graphical representation of it's space, and should have an option to convert to NTFS. I may be wrong tho.

it would clear the hard drive! so backup the whole thing, convert it and then restore
