Superman is the only super hero who was born with his powers. All other super heros had something happen to them to get their powers. Superman's alter ego is a human, where as the others are human and their alter egos are the heros. (Bruce Wayne=Batman)

Other heros are born with thier abilities: technically, Wonder Woman had them within her when she was created by her mother. While it's true that for the most part of D.C. history humans weren't born with abilities, that sort of changed after Invasion:Earth after which it became known that humans had a 'meta-human' gene (basically a bite off the mutant gene). Maybe they don't manifest until something happens to them, but they're still there.

What makes Superman stand out is this: he is the 'Pinnochio' of the super-hero genre, the wodden boy dreaming of becoming a real one. 'Kill Bill' had it wrong: he's not Superman under his Clark Kent persona, he's Kal-El. And Kal wants to fit in something fierce, if you've read the comic book. Part of it's from necessity (growing old, babies, etc.), part because he has never enjoyed the role of outsider much. He fears what his abilities will do if left unchecked, he fears his longevity... basically, he fears everything that makes him more than human.

But at the same time, he does have limitations and knows this, yet continues to risk life and limb to save those he can never be a real part of. This is 'nuture over nature', as his adoptive father Johnathan instilled these values in him. He had the best of all possible American upbringings, which makes him the D.C. equivilent of Captain America in terms of morality and patriotism. Even when the government does something stupid, he still has faith in the system in general. Go figure.

other heroes often doubt thier place in the world they protect; Superman never doubts that, but he does doubt himself. That's a big difference.

He's an alien for starters. He also has the worst disguise ever. I don't think combing your hair, taking off your glasses and wearing your undies on the outside of your tights constitutes a disguise. I think he's the only perverted super hero as well. I mean he has x-ray vision, what a perv. Other than that, he's the same as the rest, standing up for good and fighting the villian.

i think it is like 99% of the earth population are dumb. often have the wrong perception of the true hero or superman. people always try to undermine him but they always fail with full of regrets.

what a fool they are?

anyway, a fool is a fool and will never get to realise their limit and potential.

He is a kryptonian. Quoting Kill Bill, Superman is Superman, whereas his alias, Clark Kent, is the disguise, Spiderman, wakes up as Peter Parker, whereas if he puts on the costume, he is spiderman, Whereas superman, when he puts on his costume, he is Clark Kent, whearas when he removes it, he is Superman.

He is an alien, most others are mutated humans
He is the first.
He is always physically Superman, Batman et al need their costume to perform most of their job.

Superman isn't from Earth. Others might not be either but it seems significant enough to qualify such a broad question.

To begin with, he's from another planet. Most of the other heroes he hangs about with are from Earth, save Martian Manhunter or, perhaps, Hawkman.

He was the first super Hero in comic's to be come popular and paved the way for other super heroes to be made cause of it. that's what's diffent.
