I do believe the first anime to ever hit America was a dubbed version of Astro Boy. This is not including the American and Japanese co-production of some shows.
As for in Japan, the very first animation was made by an unknown creator and was an incomplete running; I think it was only around 50 frames long. Hakujaden was the first anime movie in Japan. As for the first anime series, it was Otogi Manga Calendar; hardcore black and white.

"The anime world has more than 4 decades. It began at the end of 1958 with an animated film by "Toei Animation", called "The legend of the White Snake."
But the true form of japanese animation as we know it these days, began in 1963 with the first commercial anime on TV: Testsuan Atom (Astroboy). The creator was Osamu Tezuka, a dentist who liked to draw on his spare times. One day he was watching Disney on tv, and he was surprised and fascinated by the huge eyes that Micky Mouse had, so he thought to himself what would happen is someone creates a human figure, but with those eyes.
That way, Tezuka sets the standart of japanese animation, the big eyes. And thats how the japanese animation as we know it, was really born."

I think it would be for Japan, Astro Boy which I believe comes from the 1950s
For the USA Im not entirely sure, I would make a guess and maybe say Speen racer.
But in the US anime didnt get very popular until the release of Dragon Ball Z. To many americans its known as there first taste of anime from japan.

To tell you the truth, I really don't know but I'll have to take a good stab at anything by Osama Tezuka. He pretty much created manga and anime in the sense we know it today. He did Astro Boy, Metropolis, Kimba, Phoenix etc.

well anime started got famous in the 60s, it was first discovered by america in WWII when american soldiers noticed that japanese soldiers had these cartoons with them (manga). after WWII it became popular in the US i think the first anime ever was "astro-boy" (something along the lines of super-man) anyways, that is all i know
