I kinda get the basics, but I have lots of questions.

Do you have to pay money to play?
Do wins or losses count toward your rankings, or is it all the individual player's stats?
Do you sign a team at the beginning of the year? Does that include your second and third string players, or can you adjust mid-year?
What if one of your players is injured?
How do you deal with playoffs (or do you)?
What about leagues where you pick a whole defense or a whole offense?
What else do I need to know?

In most leagues, theres a draft, it maybe live or not and you pick the predetermined number of players for your team and to fill the postions for your team, picked by the league comish before the draft.
You pick all your players in the draft, your first and second string guys can both be good players because of bye weeks, and injurys, if your player gets injured you can cut him and sign a free agent or bench him and pick up another player for the same postion. Like I had McNabb as my QB and when he tore his ACL I cut him and picked up Tony Romo.

The season in Fantasy football goes typicaly from Week 1-14. Then 15 and 16 are the playoffs.

Some leagues you pick defenseive players, or a team defense or none, its all on how the league comish sets it up.

You gain points on what your players do in the game. A teams W/L record doesnt effect your team, just the players you had active for that week matter, so if you had Warner as a benched QB for the week and he fumbled and lost it 6x you wouldnt loose the points from those fumbles, unless he was active. First off - every league has its own variation of rules and point systems. I play on Yahoo, I'll tell you how mine works:

I pay $25 to the commissioner (this is only because the winner gets a payout, I'm pretty sure the service itself is free)
Each week you are matched up against another person, whoever has the most points wins. The person with the most wins at the end of the season is the winner.
You have a draft (ours is live). You go through and pick your players. My league plays with 1 QB, 2 RB, 3 WR, 1 TE, 1 Kicker, an entire Defensive team, 1 DL, 1 DB, 1 D and 3 alternate players (which can be anything you want).
Each week you pick the players that you want on the field and on the bench, then you just watch and see what happens.
If a player gets injured, you can trade him (or keep him if he's good and you expect a quick recovery).
You can trade players with other people in the league, or you can pick people up off the Free Agent list.
Our entire Defense is scored on how many points they allow....

That's about it. It's not real hard, you just have to be good at projecting how you think players are going to play.

It depends on the league

you dont necessarily need money but you might
your player's performances turn into points and wins and losses are recorded based on who gets more points
You do a fantasy draft at the beginning of the year, but if you are in a keeper leauge after the first year you can keep some of your players from last year (and yes, you also draft 2nd and 3rd string)
Bench or cut a player who is injured, and resign a new player if you cut them
Usually the top 4 teams make the playoffs and they work just the same as regular weeks, but if you lose you are eliminated
Most leagues you draft a whole defense but offensive players come by the singles

There are pay leagues, but plenty of free ones (Yahoo, espn, etc)
Only stats count in leagues I know.
draft a team at the beginning and ytou can add or drop players all year.
If someone is hurt, put him as a reserve or if he's on IR, drop him and add another. If he's out for two or three or more, then you have to decide if he's worth keeping.
there are playoff leagues, but regular leagues have their playoffs in the last few weeks of the regular season.
Standard leagues you pick a team defense, custom you may pick individual defensive players. Don't know about whole offenses.
Try and find a custom league that looks like is at your level or better with active managers. It's no fun when everybody quits half way through in most public leagues. Don't take it too seriously. There are lots of different types of leagues, so start with one and switch to another if you don't like.

Some cost money some are free.

You draft a roster at the start of a season.

Playoffs don't count.

Scoring is based on your league settings as is your lineup.

You do get wins and losses which will allow you to make you playoff which run parrallel to an nfls week 14-17.
