If we limit ourselves to the United States, the answer is still yes.
NFL teams move when they get a better stadium deal or don't get the attendance they want. Is Dallas a major city? Guess what, the Cowboys don't actually play in Dallas.
Houston is the 4th largest city in the USA. The Oilers left for Tennessee, and it was several years before they got the Texans. The city continued to grow without an NFL team.
Look at LA and San Francisco. The teams there move from one suburb to another, and to and from the cities themselves. I would say they are actually NOT in the big cities they are near more often than they are in those cities.
The question itself tends to show one unusual feature of sports franchises: the passions they engender give them some special entitlement to public funds. The public does not think much about spending tax dollars to subsidize a sports franchise, which they would never do with say ... a meat packing plant, even though both businesses may be of similar size. Yes, having them increases tourism and recreational spending, and there is a certain civic pride in having those teams -- but in no way do they define what a major city is. In some cases, they are the biggest businesses in some small suburban towns. In other cases they are akin to a dress accessory such as a nice wristwatch (on a man), to a major city; but they do not make major cities what they are any more than they turn their sometimes small-town homes into major cities.

there's really no definition of "major city", so you're asking for an opinion.
usually "major city" means the most populous city in a state. So Seattle is the major city of Washington. But on the world scale, it barely blips.
If NFL team was a requirement to be a major city, technically Boston would not be a major city, because the Patriots are actually located about 20 miles out in Foxboro.

There is a whole world outside the USA and football... check it out sometime.
You will find that the vast majority of major cities around the world do not have a NFL team, or indeed any sort of football team whatsoever.
In fact, if you wanted to define "major city" in terms of NFL football teams for some obscure reason, it would be more accurate to state that a major city does NOT have a NFL team as only a tiny minority located in just one of the World's 190+ countries do have one.

Who is classifying? You? People can make whatever distinction they want. It's not like there is some official guidebook for when a city becomes "major."
The US Census Bureau is about as "official" as you can get and the y definitely don't care what sports teams a city has.

The answer is obviously yes. LA is the second biggest city in the country, and San Antonio is in the top ten (as far as population).
And obviously Green Bay is not a major city.
